Actually no, knowledge to the anime wouldn't necessarily be required to be able to enjoy the game. Although it will help you understand what most of the community talks about and many parts of the game environments. I am sure you'll get a bit of understanding about the anime while playing the game.

It considerable to note that once you start a game without prior knowledge of what the game is based on; you will be tempted to learn more about it, and therefore; while it would not be required to learn or know about Naruto- you will inevitably be learning via manga or the anime. That is, IF the game is of relevent interest.

You can also easily learn the main Naruto lore by playing the games. Of course, they aren't as in-depth as the Anime/Manga, but it's a good way to get an idea of what Naruto is made of.

Actually, you can. UNS and UNS2 should help a lot in understanding Naruto.

Yeah, fun fact. More people catch up on their Naruto mainly on the Storm series than the anime itself.

Anyways, aside from the Jinchuuriki and Kekkai Genkai system, you should be able to easily understand whats going on. The rank system might be complex to understand too.

Kekkai Genkai (AKA: Bloodline) is (usually) a clan of Ninja that possess a unique ability that only those who share their bloodline can do.

Jinchuuriki is somebody that had a powerful demon sealed inside of them when they were young. Only a young body (baby usually) can handle the stress of a demons chakra since the childs body is still developing.

Ranking:

Academy Student - Lowest rank. Students learning to be ninja.

Genin - Student that just graduated from the academy. They are placed in teams of 3 under a Jounin teacher to help develop their skills and teamwork.

Chuunin - Rank above Genin. Obtained by passing the Chuunin exams. They obtain their village vest to wear.

Special Jounin - A powerful ninja that had mastered at least one form of combat.

Jounin - A powerful ninja that is well rounded. Ranked ABOVE a Special Jounin

Elite Jounin - Super OP and dedicated ninja. Good at everything.

Kage - Leader of the village and most powerful ninja that lives there.

Problem solved.
